I want to move my EC2 instance, which runs on Linux Red Hat to develop a web service(using ruby/sinatra/sqs). Can anyone tell if I should use Amazon Linux on EC2 or Red Hat?

It’s better to go with Amazon Linux EC2. Because the biggest benefit to use Amazon’s Linux AMI is their pvgrub boot loader. It’s closely related to Centos, and Red Hat Linux. It also helps to compile and reboot into new or custom kernels.

With AWS Linux, one can easily implement patches being applied on boot, default AWS cmd tools, and pre-configured yum repositories.
